EEU Specification for HPLC (High Performance Liquid Chromatography)
Specification for HPLC (High Performance Liquid Chromatography) based test set to carry out Furan Analysis of insulating oils as per IEC 61198-93 and also as per ASTM-5837-99.
1. HPLC System
Fully computerized, comprehensively computer controlled HPLC system with following specifications:
a) Pump:
(i) System: Binary Gradient System, preferably with high pressure mixing, for two Mobile phase solvents: First Solvent - Water, Second Solvent - Methanol/Acetonitryl
(ii) Programming and Control: Through software running on controlling computer as well as local keyboard and display.
(iii) Programmable Flow Range: 0.01 - 5.00 ml/min with 0.01 ml increments.
(iv) Flow precision: minimum of 0.3% RSD
(v) Operating Pressure: Up to 5800 psi across entire flow Range.
(vi) Solvent compressibility: Shall be Programmable through software and it shall be possible to time-program the composition during run.
(vii) Composition Precision: <0.5% RSD
(viii) On Board diagnostics.
(ix) Various essential accessories like solvent suction / delivery tubing, micro filters, Tool kit, start-up/installation kit etc
b) Detector:
Photo Diode Array Detector of following specifications:
(i) Programmable with Built in computer Interface for data communication and control.
(ii) Minimum 512 element single stage Diode Array.
(iii) Measuring Range of 190 to 700 nm.
(iv) Accuracy: - +/- 1 nm wavelength.
(v) Light Source: - Pre-aligned D2 (Deuterium Lamp) or Simultaneous Deuterium with tungsten lamps, minimum 1000 Hrs life
(vi) Spectral Resolution / Wavelength Settability: 1.0 nm
(vii) Linear Absorbance Range: 0 to 1.5 AUFS
(viii) Base line Noise: - max ±1.5 e-5 A.
(ix) Baseline Drift: max: 0.002 AU/hour at 254 nm.
(x) Flow Cell: Quartz, 10 mm path Length, 7-15ul Volume, minimum withstand pressure-500psi.
(xi) Validation standards for verification of wavelength accuracy
(xii) Validation: Traceability certificate of calibration of source shall be provided.
(xiii) Additional Inputs / Outputs: Additional Inputs / Outputs like Analog Output, programmable event outputs, Inject start inputs etc shall be available.
c) Controlling Software
(i) Platform and interface: Software shall be common MS-Windows-7/XP/ 2000 / NT based user friendly, menu driven GUI type.
(ii) Instrument Control: Software shall be able to control entire HPLC system including pump, detector, column heating oven etc comprehensively. It shall have following features:
1. It shall be possible to carry out complete setting of parameters through software itself.
2. It shall monitor all the system components in real time and give a visual indication on the main screen that the system is functioning optimally.
3. It shall be possible to programme run-time events like, change in solvent ratios, solvent flow rates etc.
4. It shall have facility to configure, store, recall and run analytical methods, which shall hold comprehensive information like event programming, detector configuration, integrator programming, calibration tables etc to carry out complete analysis automatically.
(iii) Data Acquisition, Analysis and Storage: The software shall be capable of acquiring the run- data from PDA detector and storing it in raw form on the hard disk. It shall have following features:
1. Automatic and manual Integration with various integration functions
2. Contour plot, 3-d view
3. Comparison of two or more run data.
4. Custom Report generation with chromatograms, spectrum analysis data printouts etc
(iv) Calibration curves: Software shall have facility to accept, store and apply multi-point calibration curves for concentrations with various curve-fitting techniques for multiple wavelengths.
(v) Instrument Calibration and Validation: The software shall have facility to carry out validate and calibrate the instrument against certified standards and apply appropriate correction factor.
(vi) Other features: It is desirable that the software has additional features like wizards for setting up methods, database for organizing, storing and retrieving chromatographic data etc.
(vii) Software Validation: The software shall be fully validated. Relevant certificates of software validation shall be supplied.
d) Manual Injector:
Rheodyne Injector for Sample Injection with the following standard fittings/requirements
(i) Stand alone Mounting Plate.
(ii) Pre wired Inject switch to initiate data collection or sensor to initiate data collection.
(iii) Continuous flow during Switching.
(iv) 316 SS material.
(v) Full loop Injection
(vi) Loop size: - 20ul (Spare 5, 50, 200ul Loops)
(vii) Hamilton Syringes - 50ul, 100ul: 2 each
e) Column Heater:
HPLC Column heating with temperature programming and control
(i) Operating range - Room Temp to 60 degree
(ii) Accuracy - more or less 1.0 degree
(iii) Temperature Precision: - more or less 0.25 degree
(iv) Capacity: - Two columns with guard columns.
(v) Control through software
f) HPLC Columns:
(a) C18, 3.9 x 250/300mm, 60A, 4um (Reversed phase column packed with Octyl or Octadecyl bonded Silica - 1 No.
(b) Guard-Column packed with the same material as the analytical column along with In line Guard Cartridge holder kit–1 No.
g) END Connector Kit:
Complete plumbing accessories for connecting solvents reservoir to pump, pump to injector, injector to guard column, guard column to main column, main column to detector and detector to drain etc.,
h) Computer with laser printer:
a) Controlling Computer shall have following minimum specifications: Intel Pentium IV@1.5 GHz, Bus speed min 133MHz, HDD - 40 GB ATA, SVGA AGP display card with 32 MB VRAM, CD-ROM Drive 48X, 512 MB RAM, 15" flat screen color monitor, 10/100MBPS LAN card and licensed OS (OS shall be the one on which the controlling software shall run).
b) However, if the HPLC system Software being offered requires higher that the above specifications, it shall be ensured that the computer offered is of sufficient specifications, as required to the offered software.
c) Printer shall be HP-laserjet-1200.
